Cy3 Labeled Negative Control siRNA

Cy3 Labeled Negative Control siRNA duplex is a 3’-dye-conjugated siRNA transfection efficiency control with no homology with mammalian genes. The synthetic siRNAs (5’-UUC UCC GAA CGU GUC ACG UTT-Cy3-3’ and 5’- ACG UGA CAC GUU CGG AGA ATT-3’) enable qualitative assessment of cellular uptake, distribution and intracellular localization of siRNAs 1-4 hours post transfection. Cy3 Negative Control siRNA is commonly used to optimize transfection conditions. Transfection can be monitored by fluorescence microscopy or FACS.

Cy3 Negative Control siRNA duplex resolves as a single band via PAGE

Lyophilized powder

Reconstitution Protocol:

Briefly centrifuge the tubes (maximum RCF 4,000g) to collect lyophilized siRNA at the bottom of the tube. Resuspend the siRNA in 25 ul of DEPC-treated water (supplied by researcher), which results in a 1x stock solution (100 uM). Gently pipet the solution 3-5 times to mix and avoid the introduction of bubbles. Optional: aliquot 1x stock solutions for storage.

Spectral Information:

Filter: Cy3/TRITC
Excitation max (lambda max): 550 nm
Emission max (lambda max): 570 nm.

Storage and Stability:

The lyophilized powder is stable for at least 4 weeks at room temperature. It is recommended that the lyophilized and resuspended siRNAs are stored at or below -20oC. After resuspension, siRNA stock solutions ≥2 uM can undergo up to 50 freeze-thaw cycles without significant degradation. For long-term storage, it is recommended that the siRNA is stored at -70oC. For most favorable performance, avoid repeated Handling and multiple freeze/thaw cycles.
