Overview:

The Parkin protein is a component of the E3 ubiquitin ligase complex, which is responsible for targeting of proteins for degradation. Hereditary Parkinson’s disease is most commonly caused by mutations in the parkin gene. The disease is characterized by the progressive loss of dopaminergic neurons and the presence of Lewy bodies in the substania nigra (1). Recent studies indicate that phosphorylation of Parkin at Ser101 and Ser378 might be important for regulating E3 ubiquitin ligase activity (2).

Stability:

Store at 4oC (add 0.1% NaN3) for several months, and at -20oC for longer periods. For optimal storage, aliquot target into smaller quantities after centrifugation and store at recommended temperature. For most favorable performance, avoid repeated handling and multiple freeze/thaw cycles.


Sample Data:

Western blot of HEK293 cells transfected with Parkin WT (Phospho) and Parkin S378 mutant (non-phospho) showing the phospho-specific immunolabeling of the ~ 52 kDa Parkin protein. The immunolabeling is absent in the Parkin S378 mutant.
