Kinases

Protein kinases are a family of pivotal enzymes with 518 members implicated in various processes of signal transduction, constituting over 2% of the human genome. Kinases regulate cell signaling pathways by phosphorylating their physiological substrates. They catalyze the transfer of γ-phosphate from ATP or GTP to the hydroxyl groups of serine, threonine and tyrosine residues on the substrate proteins or the glycosyl moieties in lipids. Kinase deregulation could lead to the changes in kinase enzymatic activity, subcellular localization, stability of kinase, protein-protein interactions, and thereby aberrant activation of downstream signaling pathways that underlie all major human pathological conditions such as cancer, cardiovascular disease, metabolic disorders, and more.
