Overview:
MYCN is a basic helix-loop-helix transcription factor first identified in neuroblastoma and is a member of the MYC proto-oncogene family along with MYCC and MYCL. MYC proteins affect transcription of many genes thus regulating proliferation, metabolism, apoptosis, differentiation and immune surveillance. Ubiquitin-dependent proteasome degradation of MYCN in neural stem/progenitor cells is required for the arrest of proliferation and the start of differentiation. Enhanced USP7 expression is seen in neuroblastoma patients with poorer prognosis due to deubiquination of MYCN by USP7, thereby preventing degradation of MYCN (1). Overexpression of MYCN is closely related with high-cancers, early distant metastasis, and poor clinical prognosis (2).
